The nuclear factor erythroid 2-related factor (Nrf2) is a key transcription factor that regulates the expression of a number of critical redox homeostatic genes. Under normal environmental conditions, Nrf2 is anchored to the actin cytoskeleton via interaction with kelch-like ECH-associated protein 1 (KEAP1). This interaction prevents Nrf2 from entering the nucleus by sequestering Nrf2 in the cytosol and also aiding in its degradation. Under instances of oxidative stress, Nrf2 is released from KEAP1 and freed to translocate to the nucleus and activate transcription of antioxidant response elements (ARE)-associated genes.
The cytoprotective genes activated by Nrf2 encode proteins that provide the cell with protection from reactive oxygen species (ROS) and a number of other dangers. The protective proteins include conjugating enzymes, antioxidant enzymes, anti-inflammatory proteins, repair proteins, and others. Given the protective role of Nrf2 gene activation, scientists are searching for therapeutics that can inhibit KEAP1:Nrf2 interaction, thereby activating ARE-associated genes. This is especially important in the case of cancer, where ROS are well-known to be a major etiological factor in disease onset and progression.
