“Tissue culture grades water” has been around for decades and is well known to every laboratory researcher involved in growing live tissue cell cultures. Summer dry eye symptoms, according to Kleyne, tend to occur when temperatures are hot, humidity is low and air pollution is high. The combination greatly increases the rate at which water evaporates from the delicate tear film covering the surface of the eye, which should be 99 percent water. The one-percent that is not water - salt, beneficial proteins, antibodies and lipids (oils) - does not evaporate and becomes overly concentrated when the water evaporates.
Kleyne’s solution for humidifying ocular surfaces affected by summer dry eye was to apply a pure all-water mist to the air in front of the eyes, allowing the eyes to naturally absorb as much airborne moisture as they need – an amount usually measured in nanoliters. In seeking an ideal water to convert to a mist to benefit dry eyes, Kleyne zeroed in on “tissue culture grade water,” also called “cell culture grade water.”
The specialized water, Kleyne explains, is normally used to reconstitute powdered cell growth medium for laboratory cell cultures. For optimal cell growth, the water must have several specifically defined qualities. Above all, the waters’ attributes must not vary or it could skew the results.
According to Julia Oxford, PhD, a molecular biologist at Boise State University, nearly 100 percent of tissue culture grade water used in laboratory research is artificially manufactured. To remove impurities, regular tap water is micro-filtered, undergoes reverse osmosis and deionization and is sterilized
Tissue-culture grade water is high grade water with no impurities or contaminants, explains Oxford. The pH of tissue-culture grade water should be slightly acidic, between 5.7 and 7.0. Inorganic minerals, essential for cell growth, must be present in minute quantities. Organic matter is absent from tissue-culture grade water. Microorganisms such as bacteria, viruses and algae are absent, as are organic breakdown products.
When Oxford tested the Trade Secret water from Bio-Logic Aqua® Research’s exclusive water source, she found that it met or exceed all criteria for tissue culture grade water. Furthermore, the analysis was more consistent than manufactured water. To Oxford’s knowledge, no all-natural water had ever before been found that meets these standards.
“I knew our Trade Secret water was special,” says Kleyne, “but I never suspected it was that special. When you think about it, though, water that is ideal for growing live human tissue cells in the laboratory should also be ideal for maintaining healthy eyes and skin.”
