GPCRs can be modified so that they only respond to a specific biologically inert chemical (a designer drug). These modified GPCRs are called Designer Receptors Exclusively Activated by Designer Drugs or “DREADDs”. Cells expressing the DREADD respond robustly to low concentrations of the designer molecule, whereas cells that do not express the DREADD are unresponsive to the designer drug. DREADDs are functionally similar to their wild type receptors, therefore ligands such as Clozapine N-oxide (CNO), that activate DREADDs are exciting tools for the study of GPCR signaling.
CNO potently activates various muscarinic DREADD receptors. These receptors have no affinity for the endogenous ligand acetylcholine (ACh), but are responsive to CNO. CNO has no significant affinity for other CNS targets.
Clozapine N-oxide joins a range of affordable and high quality tools for GPCR research, including the novel GRK2/3 inhibitor Cmpd101, that can be used to study GPCR desensitization. Hello Bio also offers standard tools such as D-AP5.
