It is well established that ligand binding can protect proteins from thermal denaturation in a cellular milieu; however, the broad application of this cellular target engagement concept to drug discovery has been hindered by a lack of simple platforms with sensitive quantitative readouts. Cell-based thermal stabilization assays are valuable methods for particular applications, but they can require target-specific antibodies for immuno-assay readouts, can be low-throughput for Western blot formats, and can be biased against multi-domain proteins.

In this webinar, participants will get an overview of compound-target engagement, and overview of the applications of InCELL Pulse™, which uses DiscoverX's Enzyme Fragment Complementation (EFC) technology to overcome these limitations of current thermal stabilization assays. This unique platform enables users to rapidly measure quantitative cellular target engagement potency values for inhibitors of diverse protein classes, including kinases, methyltransferases, and hydrolases.

Also covered in this webinar will be the tools and workflow necessary to design InCELL Pulse assays, as well as validation data. Dr. Dana Haley-Vicente, senior product manager with the DiscoverX Corporation, has been selected to be the speaker for this event.

Haley-Vicente earned a doctorate in pharmacology from the University of California, Los Angeles and has more than 16 years of experience working in the life science drug discovery industry. Currently she is a senior product manager at DiscoverX, where she manages cell-based assays, kits, and consumable products for drug discovery target discovery, screening, lead optimization, and SAR campaigns.
