This new IQbot provides preclinical researchers exploring angiogenesis in wound healing and cancer treatments with the ability to automate their quantitation procedure while eliminating the imprecision and effort of manual tube formation image analysis and expanding the amount of data available – all online. “IQbots” are the engines that power the ImageQuantify.com analysis and management workflow — tools for common or custom preclinical imaging assays that deliver validated, comprehensive analytics that are objective, reproducible, and accessible. Within every IQbot results package, researchers receive complete data (i.e., quantitative excel spreadsheets, image analysis overlays, usable file formats), image analysis validation certificate, and reference materials (e.g., a video walkthrough of output data, relevant references from scientific literature, materials/methods section(s), and material to support GLP compliance).
Endothelial cells, when provided with a 3D extracellular matrix substrate and appropriately supplemented growth media, will self-organize into a network of capillary-like "tubes." This in-vitro assay is commonly used to assess the ability of compounds to stimulate or inhibit angiogenesis. To perform the assay, the endothelial cells are usually seeded (subconfluently) in multi-well plates filled with matrigel (extracellular matrix). While not absolutely necessary (phase-contrast imaging is sufficient although often corrupted by uneven lighting due to the meniscus formed in each well), endothelial cell membranes may be labeled with fluorescent DiI dyes added directly to each well. Each well is subsequently imaged using a high resolution, wide-field microscope using both phase-contrast and fluorescence modes. Images are acquired while raster scanning across each well to form a single, high-resolution mosaic (i.e., multi-tile) image. To analyze a given image, the IQbot applies a series of custom morphological and spectral filters to the mosaic, allowing the endothelial cell network to be segmented and skeletonized for nodal and branching analyses. In doing so, the IQbot is able to report specific area, lumen, node, and branch characteristics of the network.
This IQbot will join the existing ImageQuantify.com catalog in assisting users to more effectively (in terms of the quality standard of the data) and more efficiently (in terms of cost and time-to-analysis) generate the imaging data for their research. As with all prior offerings, the Tube Formation Analysis IQbot was developed and released following a rigorous validation process that stressed the accuracy and reproducibility of the produced data.
